目的:观察大黄素对精索静脉曲张缺氧模型精原细胞的保护作用。方法:选用小鼠GC-1精原细胞,设置对照组(A组)、对照+大黄素组(B组)、模型组(C组)、模型+大黄素组(D组)。C组和D组加入氯化钴(CoCln 2)至终质量浓度为100 μmol/L,然后培养细胞24 h构建精索静脉曲张缺氧模型。B组和C组加入大黄素至终质量浓度为40 μmol/L。采用细胞计数试剂-8(CCK-8)法及流式细胞仪测定细胞凋亡水平,采用蛋白质印迹法(Western blot)检测半胱氨酰天冬氨酸特异性蛋白酶(Caspase-3)和B细胞淋巴瘤/白血病-2相关X蛋白(bax)的蛋白质表达水平,采用实时定量反转录聚合酶链式反应(RT-qPCR)检测细胞Caspace-3和bax的基因表达水平,组间比较采取n t检验。n 结果:CCK-8实验示C组和D组细胞存活率低于A组[(51.70±3.52)%、(77.84±5.18)%比(100.00±0.00)%,n t=23.767、7.410,n P值均<0.05],差异有统计学意义,且D组细胞存活率高于C组[(77.84±5.18)%比(51.70±3.52)%,n t=7.229,n P<0.05],差异有统计学意义。流式实验示D组凋亡细胞比例低于C组[(10.54±0.68)%比(21.23±1.01)%,n t=15.207,n P<0.05],差异有统计学意义。Western blot结果示D组Caspace-3和bax蛋白表达水平低于C组(0.713±0.020、0.819±0.022比1.134±0.025、1.271±0.019,n t=22.722、26.813,n P值均<0.05),差异有统计学意义。RT-qPCR结果示D组Caspace-3和bax表达水平低于C组(1.52±0.18、1.80±0.21比3.53±0.23、4.27±0.31,n t=11.920、11.426,n P值均<0.05),差异有统计学意义。n 结论:大黄素通过抑制细胞凋亡对精索静脉曲张缺氧模型的精原细胞起保护作用。“,”Objective:To investigate the efficacy of emodin protecting spermatogonia from CoCln 2-induced hypoxia in varicocele model.n Methods:Mouse GC-1 cells were selected and four groups were set up as control group (group A), emodin control group (group B), hypoxia group (group C), hypoxia+ emodin group (group D). In group C and group D, CoCln 2 was added to the final concentration of 100 μmol/L, and cells were cultured for 24 hto estabilish hypoxia model. In group B and group C, emodin was added to the final concentration of 40 μmol/L. The level of apoptosis was detected by cell counting kit-8 (CCK-8) assay and flow cytometry. Western blotting and real-time quantitative reverse transcriptase-polymerase chain reaction (RT-qPCR) were used to detect the expression of B cell lymphoma/leukemia-2 associated X protein (bax) and cysteinyl aspartate-specific protease (Caspase)-3.n Results:Compared with group A, the survival rate was decreased in group C (51.70±3.52)% and group D (77.84±5.18)%. The survival rate in group D was increased as compared with group C (n t=7.229, n P<0.05). The apoptosis rate in group D (10.54±0.68)% was decreased as compared with group C (21.23±1.01)%,n t=15.207, n P<0.05). The expression level of Caspase-3 and bax in group D (1.134±0.025, 1.271±0.019; 1.52±0.18, 1.80±0.21) was decreased as compared with group C (0.713±0.020, 0.819±0.022; 3.53±0.23 and 4.27±0.31) in Western blotting (n t=22.722, 26.813, n P<0.05) and RT-qPCR (n t=11.920, 11.426, n P<0.05).n Conclusion:Emodin protects spermatogonia from CoCln 2-induced hypoxia by inhibiting apoptosis in varicocele model.n
