目的:从小牛胸腺中提取胸腺素并检测药物指标,观察其对哮喘豚鼠肺组织 Gqα表达的影响。方法:酸碱沉淀加离心法制备胸腺素,经多肽定量、活性鉴定、细菌培养、毒性试验和热原实验检测其药物指标。卵蛋白雾化吸入致敏法复制哮喘模型, Western blot(蛋白免疫印迹 )分析法测定 Gqα水平。结果:所制胸腺素符合中国药典标准。其治疗哮喘豚鼠后,肺组织 Gqα水平显著降低 (P<0.5)。结论:采用酸碱沉淀加离心法可制备胸腺素,其治疗哮喘的机理可能是通过改变肺组织Gqα水平来达到。
OBJECTIVE: To extract thymosin from calf thymus and measure the index of the drug to observe its effect on the expression of Gqα in lung tissue of asthmatic guinea pigs. Methods: Thymosin was prepared by acid-base precipitation and centrifugation. The drug index of the thymosin was determined by quantitative analysis of the polypeptide, activity identification, bacterial culture, toxicity test and pyrogen test. Asthmatic model was induced by inhalation of ovalbumin, and Gqα level was determined by Western blot analysis. Results: The prepared thymosin in line with the Chinese Pharmacopoeia standards. After treatment of asthmatic guinea pigs, the level of Gqα in lung tissue was significantly decreased (P <0.5). Conclusion: Thymosin can be prepared by acid-base precipitation and centrifugation. The mechanism of asthma treatment may be achieved by changing the level of Gqα in lung tissue.
