目的 观察高压氧 (HBO)、脑内注射脑源性神经营养因子 (BDNF)联合治疗新生大鼠缺氧缺血性脑损伤 (HIBI)的疗效。方法 7d龄 SD大鼠左侧颈总动脉结扎联合低氧吸入形成 HIBI后 ,即刻脑室注射 BDNF(0 .5 μg)并行 HBO治疗 (A)或常氧高压氮处理 (B) ,设单纯 HIBI组 (C)、假手术组(D)和正常组 (E) ,观察对脑水肿、皮层和海马脂质过氧化物 (MDA)水平和细胞凋亡百分率的影响。结果 A,B组脑水肿程度、MDA和细胞凋亡百分率水平均显著低于 C组 ;脑水肿程度、细胞凋亡百分率A组显著低于 B组 ,尤以脑水肿降低更明显 ,接近于 D组 ,MDA水平两组相差无显著意义。结论 联合应用 HBO及脑室注射 BDNF治疗可显著减轻 HIBI后的脑损伤 ,其效果优于 BDNF+常氧高压氮处理组 ,表明在 HIBI的治疗中 HBO与 BDNF联合应用起到了协同作用。
Objective To observe the effects of hyperbaric oxygen (HBO) and brain injection of brain-derived neurotrophic factor (BDNF) on neonatal hypoxic-ischemic brain damage (HIBI). Methods The left common carotid arteries of 7-day-old SD rats were ligated with hypoxia inhalation to form HIBI. Immediate intracerebroventricular injection of BDNF (0.5 μg) followed by HBO treatment (A) or normoxia nitrogen treatment (B) (C), sham-operation group (D) and normal group (E). The effects on cerebral edema, cortical and hippocampal lipid peroxide (MDA) levels and percentage of apoptosis were observed. Results The degree of brain edema and the percentage of MDA and apoptosis in group A and group B were significantly lower than those in group C; The degree of cerebral edema and the percentage of apoptosis in group A and group B were significantly lower than those in group B, especially in brain edema There was no significant difference between the two groups in the levels of MDA and MDA. Conclusions Combined use of HBO and intraventricular injection of BDNF can significantly reduce brain injury after HIBI, and its effect is better than that of BDNF + normoxic high-pressure nitrogen treatment group, indicating that synergistic effect of HBO and BDNF in HIBI treatment is synergistic.
