目的:比较5种试验/探针系统检测感染兔血清中循环抗原(SCA)的效果。方法:用单雄性和单雌性尾蚴分别感染9只家兔,以复性尾蚴感染7只家兔作对照;感染后定期采血。另15兔分3组感染250条尾蚴。1,2组于感染后19d和45d分别开始给予丙烯基硫脲(295-590mg/kg)以抑制虫卵形成,第3组作对照;均定期采血。检测方法计5种:1斑点-ELISA/抗成虫表膜单抗(8SE4),2斑点-ELISA/抗CCA单抗(3D10),3夹心斑点-ELISA/抗虫卵单抗(MG2),4夹心法-ELISA/抗虫卵单抗(2H10),5夹心法-ELISA/抗CAA单抗(IB10)。结果:9只单雄性尾蚴和9只单雌性尾蚴感染兔血清1法均为阴性,2法仅1只检出雄虫133条的家兔出现阳性反应,5法单雌性感染均阴性、单雄性感染3/9兔阳性。服药抑制虫卵形成的兔血清,19d开始服药5兔1法及3法均未检出SCA,46d开始服药组在感染后6-7wk二法均检出SCA。4法服药与未服药兔均为阴性,5法感染后6wk的血清服药与未服药兔均呈阳性反应。结论:不同的试验/探针系统的检测效果不同,但所试3种方法(1,2,5种)都不能检出单雌性感染。抑制虫卵形成?
OBJECTIVE: To compare the effects of five test / probe systems on the detection of circulating antigens (SCA) in infected rabbits. Methods: Nine rabbits were infected with single male and female respectively, and seven rabbits infected with cercariae were used as controls. Blood samples were taken periodically after infection. Another 15 rabbits divided into 3 groups infected 250 cercariae. Group 1 and group 2 received propanethiourea (295-590 mg / kg) at 19d and 45d post-infection, respectively, to inhibit egg formation. Group 3 served as control; blood was collected routinely. Five detection methods were as follows: 1 spot-ELISA / anti-adult membrane monoclonal antibody (8SE4), 2 spot-ELISA / anti-CCA monoclonal antibody (3D10), 3 sandwich spot-ELISA / Sandwich method - ELISA / pestobolomycin (2H10), 5 Sandwich method - ELISA / anti-CAA monoclonal antibody (IB10). Results: Nine rabbits with single cercariae and nine single-cercaria infected rabbits were negative in serum 1 test. In the 2 rabbits, only one of 133 rabbits tested positive for the disease. Five single-female infectives were all negative and single-male Infection 3/9 rabbit positive.
