目的研究Hp感染对胃黏膜FOXP3、TGF-β1、IL-10表达的影响。方法采用原位杂交和ELISA法检测Hp感染与未感染患者胃黏膜活检组织中FOXP3m RNA、TGF-β1m RNA的表达和IL-10的含量;用ELISA法检测胃黏膜上皮细胞GES-1与Hp共培养后的TGF-β1、IL-10含量。结果 (1)Hp感染阳性患者胃黏膜组织中FOXP3m RNA、TGF-β1m RNA的阳性表达率及IL-10的含量较Hp感染阴性患者明显增高(分别为P<0.05、P<0.05和P<0.01);(2)Hp感染阳性患者胃黏膜组织FOXP3m RNA的表达与TGF-β1m RNA和IL-10的表达呈正相关(分别为P<0.05和P<0.01);(3)胃黏膜上皮细胞GES-1与Hp共培养1h后,TGF-β1及IL-10的表达量均较阴性对照组明显增高(P<0.05)。结论 Hp感染可上调胃黏膜局部CD4+CD25+FOXP3+Treg细胞,诱导胃黏膜细胞分泌TGF-β和IL-10,从而抑制胃粘膜局部免疫,促进Hp的免疫逃逸和持续性感染。
Objective To study the effect of Hp infection on the expression of FOXP3, TGF-β1 and IL-10 in gastric mucosa. Methods The expression of FOXP3mRNA and TGF-β1mRNA and the content of IL-10 in gastric mucosal biopsies from patients with or without Hp infection were detected by in situ hybridization and ELISA. The expressions of GES-1 and Hp in gastric mucosal epithelial cells After training TGF-β1, IL-10 content. Results (1) The positive rates of FOXP3m RNA and TGF-β1mRNA in gastric mucosa with positive Hp infection and the levels of IL-10 in gastric mucosa were significantly higher than those with negative Hp infection (P <0.05, P <0.05 and P <0.01, respectively ). (2) The expression of FOXP3mRNA in gastric mucosa with positive H.pylori infection was positively correlated with the expression of TGF-β1m RNA and IL-10 (P <0.05 and P <0.01 respectively). (3) The expression of FOXP3mRNA in gastric mucosal epithelial cells, GES- 1 and Hp co-cultured for 1 h, TGF-β1 and IL-10 expression were significantly higher than the negative control group (P <0.05). Conclusions Hp infection can upregulate the local CD4 + CD25 + FOXP3 + Treg cells in gastric mucosa and induce the secretion of TGF-β and IL-10 in gastric mucosa cells, thus inhibiting the local immunity of gastric mucosa and promoting the immune escape and persistent infection of Hp.
