肿瘤光动力治疗 (photodynamictherapy ,PDT)是现代癌症治疗中一种新的治疗法 ,为了更好地认识光动力诱导肿瘤细胞凋亡的作用机制。方法 :用Fluo 3AM荧光探针负载培养的人肺腺癌GLC 82细胞 ,在激光扫描共聚焦显微镜下观察细胞内Ca2 +浓度的变化。结果 :激光单独照射前后GLC 82细胞内Ca2 +的浓度变化不大 ;在PDT作用下 ,激光照射前GLC 82细胞内Ca2 +的浓度变化明显。结论 :本方法灵敏、简单、快速 ,能实时监测细胞内游离钙的变化。
Photodynamic therapy (PDT) is a new treatment method in modern cancer treatment, in order to better understand the photodynamic therapy-induced apoptosis of tumor cells. METHODS: Human lung adenocarcinoma GLC 82 cells were cultured with Fluo 3AM fluorescent probe and the changes of intracellular Ca2 + concentration were observed under a laser scanning confocal microscope. RESULTS : The concentration of Ca2 + in GLC 82 cells before and after laser irradiation alone had little change. Under PDT, the concentration of Ca2 + in GLC 82 cells before laser irradiation changed significantly. Conclusion : This method is sensitive, simple and rapid. It can monitor the changes of intracellular free calcium in real time.
