目的:研究护肝布祖热胶囊原料药制备工艺。方法:水蒸气蒸馏法提取芹菜子、小茴香中挥发油,在单因素试验的基础上,以挥发油量为指标,正交试验法确定出挥发油提取最佳工艺和挥发油β-CD包合工艺。以秦皮乙素含量和醇提取物得率作为评价指标,正交试验考察乙醇提取复方其余药材的最佳工艺。结果:确定出挥发油提取工艺为药材加入12倍量水,浸泡2 h后提取8 h。挥发油最佳包合工艺为挥发油与β-CD比例为1∶6,包合温度40℃,包合时间1 h。乙醇提取最佳工艺为10倍量50%的乙醇提取3次,每次2 h。结论:优选得到的提取工艺合理、可行,将挥发油包合物、水提取物和乙醇提取物充分混匀作为胶囊原料药。
Objective: To study the preparation technology of Huganbuzu heat capsule raw material. Methods: Volatile oil was extracted from celery seeds and cumin by steam distillation. Based on the single factor experiment, volatile oil content was taken as an index. The optimum extraction process of volatile oil and β-CD inclusion process were determined by orthogonal test. Aesculin content and alcohol extract yield as an evaluation index, the orthogonal test ethanol extraction compound herbs the best technology. Results: The essential oil extraction process was determined to be 12 times the amount of water added to the medicinal materials, soaked for 2 h and then extracted for 8 h. Volatile oil best inclusion process for the volatile oil and β-CD ratio of 1: 6, inclusion temperature 40 ℃, inclusion time 1 h. The best ethanol extraction process for 10 times the amount of 50% ethanol extraction 3 times, each 2 h. Conclusion: The optimal extraction process is reasonable and feasible, and the volatile oil inclusion compound, water extract and ethanol extract are fully mixed as capsule drug substance.
