目的应用代谢组学方法筛选中年2型糖尿病患者尿液中的小分子生物标志。方法采用液相色谱-质谱联用技术(LC-MS)检测中年2型糖尿病患者和对照对象尿样中的内源性代谢产物,对检测所得总离子流图进行特征离子峰提取(MFE)、滤噪、峰匹配、峰对齐、归一化及基线校正,经t检验对在两组研究对象间有统计学差异(P<0.05)的化合物筛选鉴定。结果筛选出3个在两组研究对象尿样中表达水平有统计学差异(P<0.05)的代谢物:色氨酸、犬尿酸和犬尿酸原。中年2型糖尿病患者体内代谢产物色氨酸、犬尿酸经尿液代谢的水平下降,犬尿酸原经尿液代谢的水平升高。结论色氨酸、犬尿酸和犬尿酸原可能是与中年2型糖尿病相关的生物标志。
Objective To screen small molecule biomarkers in urine of middle-aged type 2 diabetics using metabonomics. Methods The endogenous metabolites in urine of middle-aged patients with type 2 diabetes mellitus and controls were detected by liquid chromatography-mass spectrometry (LC-MS), and the characteristic ion peak extraction (MFE) , Noise filtering, peak matching, peak alignment, normalization and baseline correction. The t test was used to screen and identify compounds with statistical difference (P <0.05) between the two groups. Results Three metabolites, tryptophan, kynurenic acid and kynurenine, were found to be significantly different (P <0.05) in urine samples from two groups. Middle-aged patients with type 2 diabetes tryptophan metabolites, kynurenal decline in the level of urine metabolism, kynurenine by the level of urine metabolism. Conclusion Tryptophan, kynurenic acid and kynurenine may be associated with middle-aged type 2 diabetes-related biomarkers.
