目的:探讨rhBMP-7对腺嘌呤致大鼠慢性肾衰竭的防治作用。方法:将Wistar大鼠随机分为正常组、模型组、模型对照组、rhBMP-7预防组、rhBMP-7小、大剂量治疗组。除正常组外,其余各组按300mg.kg-1.d-1用腺嘌呤灌胃,rhBMP-7预防组在灌胃的同时按300μg.kg-1隔日腹腔注射rhBMP-7。在d 35各组停止灌胃并处死正常组、模型组、rhBMP-7预防组的大鼠,取血,作生化指标及血液指标检测,并留取肾组织。从d 36开始,模型对照组按1 mL.kg-1的剂量隔日腹腔注射生理氯化钠溶液,rhBMP-7小、大剂量治疗组分别以100和300μg.kg-1隔日腹腔注射rhBMP-7。在d 70处死大鼠,取血,作生化指标及血液指标检测,并留取肾组织。结果:rhBMP-7在不同程度上降低慢性肾衰大鼠的BUN,Cr,UA,磷含量,增加钙含量,改善贫血,降低肾脏指数,减轻肾衰大鼠的肾脏损伤。结论:rhBMP-7对腺嘌呤致大鼠慢性肾衰竭有一定的防治作用,为探索治疗肾功能衰竭提供新的途径。
Objective: To investigate the preventive and therapeutic effects of rhBMP-7 on chronic renal failure induced by adenine in rats. Methods: Wistar rats were randomly divided into normal group, model group, model control group, rhBMP-7 prevention group, rhBMP-7 small dose and high dose treatment group. In addition to the normal group, the other groups were given gavage with adenine at 300mg.kg-1.d-1, and intraperitoneal injection of rhBMP-7 at 300μg.kg-1 in rhBMP-7 prophylaxis group. Rats in the normal control group, model group and rhBMP-7 prevention group were given gavage and sacrificed at d 35, and blood was taken for biochemical and blood tests. Renal tissues were collected. Starting from d 36, the model control group was intraperitoneally injected with physiological sodium chloride solution at a dose of 1 mL.kg-1, while rhBMP-7 was given to small dose and high dose treatment groups at the intervals of 100 and 300 μg.kg-1 . Rats were sacrificed at d 70, blood was taken for biochemical and blood test, and renal tissue was collected. Results: rhBMP-7 reduced BUN, Cr, UA, P content, increased calcium content, improved anemia, decreased renal index and renal damage in rats with chronic renal failure to varying degrees. Conclusion: rhBMP-7 can prevent and cure chronic renal failure induced by adenine in rats, and provide a new way to explore the treatment of renal failure.
