目的研究大鼠肠道菌群对没食子酸和原儿茶酸2个单体成分的代谢作用。方法建立UHPLC-Q-TOF/MS检测条件,流动相为体积分数0.1%甲酸水溶液(A)-体积分数0.1%甲酸乙腈溶液(B)梯度洗脱,流速为0.3 m L·min-1,扫描方式为电喷雾(ESI)负离子模式。采用负离子和多反应离子监测模式分析没食子酸和原儿茶酸分别与大鼠肠道菌群共孵育后的代谢产物。结果从药物菌群孵育液中鉴定出没食子酸和原儿茶酸在大鼠肠道细菌各种酶的作用下发生脱羧和甲基化反应。结论阐明了没食子酸和原儿茶酸在大鼠肠道菌群中的代谢特征。
Objective To study the metabolism of two monomer components of gallic acid and protocatechuic acid in intestinal flora of rats. Methods The detection conditions of UHPLC-Q-TOF / MS were established. The mobile phase consisted of gradient elution with a volume fraction of 0.1% formic acid in water (A) - 0.1% formic acid in acetonitrile and a flow rate of 0.3 m L · min- The mode is electrospray (ESI) negative ion mode. The negative ion and multi-reactive ion monitoring modes were used to analyze the metabolites of gallic acid and protocatechuic acid co-incubated with intestinal flora in rats. RESULTS: Gallic acid and protocatechuic acid were identified from the drug-flora incubation solution. The decarboxylation and methylation reaction occurred under the action of various enzymes in intestinal bacteria of rats. Conclusion The metabolic characteristics of gallic acid and protocatechuic acid in rat intestinal flora were clarified.
