目的探讨天麻对帕金森病模型鼠黑质(SN)和腹侧被盖区(VTA)TNF-α及胶质源性神经营养因子(GDNF)表达的影响。方法 Wistar大鼠60只,随机分为正常组、模型组、美多巴组与大、中、小剂量天麻组,每组10只,采用免疫组织化学ABC法分别进行TNF-α和GDNF阳性细胞表达的测定。结果与正常组比较,大剂量天麻组和模型组左侧SN、VTA中TNF-α表达明显升高(P<0.05,P<0.01);除小剂量天麻组左侧SN、VTA中GDNF表达明显升高(P<0.05),其余各组GNDF表达差异无统计学意义(P>0.05);与模型组比较,美多巴组、中、小剂量天麻组左侧SN中TNF-α表达明显降低,小剂量天麻组左侧VTA中TNF-α表达明显降低(P<0.05),中、小剂量天麻组左侧SN、VTA中GDNF表达明显升高(P<0.05);与美多巴组比较,小剂量天麻组VTA中的GDNF表达亦明显升高(P<0.05)。结论在6-羟基多巴胺毁损纹状体模型中,小剂量天麻显著下调TNF-α的表达和上调GDNF的表达,可能是天麻神经免疫调节的机制之一。
Objective To investigate the effect of Gastrodia on the expression of TNF-α and GDNF in substantia nigra (SN) and ventral tegular area (VTA) of Parkinson’s disease model rats. Methods Sixty Wistar rats were randomly divided into three groups: normal group, model group, metoprolol group and large, medium and small doses of Tianma group, with 10 in each group. Immunohistochemical ABC method was used to detect the expression of TNF-α and GDNF positive cells Determination of expression. Results Compared with the normal group, the expression of TNF-α in SN and VTA on the left side of the high-dose gastrodia tuber and model group was significantly increased (P <0.05, P <0.01) (P <0.05). There was no significant difference in the expression of GNDF between the other groups (P> 0.05). Compared with the model group, the expression of TNF-α in left SN of gastropod group was significantly decreased (P <0.05). The expression of GDNF in SN and VTA of middle and low dose Tianma group was significantly increased (P <0.05). Compared with the model group, the expression of TNF- The GDNF expression in VTA of low-dose gastrodia tuberosus group was also significantly increased (P <0.05). Conclusion In the 6-hydroxydopamine-lesioned striatum model, the low dose of Gastrodia elata significantly down-regulated the expression of TNF-α and increased the expression of GDNF, which may be one of the mechanisms of neuroimmunity regulation in Gastrodia elata Blume.
