目的探讨黄芪多糖对急性脊髓损伤转化生长因子(TGF-β1)表达的影响。方法参照Nystrom压迫方法制作大鼠脊髓压迫损伤模型,成年健康Wistar大鼠72只,雌雄不限,随机分为假手术组、损伤组和黄芪多糖组,每组32只。免疫组化方法和Western blot方法检测各组大鼠TGF-β1的表达。结果免疫组化结果显示,脊髓损伤组与黄芪多糖组大鼠TGF-β1的表达显著低于假手术组,而与脊髓损伤组相比,黄芪多糖组大鼠TGF-Β1的表达明显升高(P<0.01);Western blot方法也得到了相同的结果。结论黄芪多糖很可通过上调TGF-β1的表达参与脊髓损伤。
Objective To investigate the effect of astragalus polysaccharides on the expression of transforming growth factor-β1 (TGF-β1) in acute spinal cord injury. Methods According to Nystrom compression method, 72 adult Wistar rats were randomly divided into sham operation group, injury group and Astragalus polysaccharide group, with 32 rats in each group. Immunohistochemistry and Western blot were used to detect the expression of TGF-β1 in each group. Results The results of immunohistochemistry showed that the expression of TGF-β1 in spinal cord injury group and astragalus polysaccharide group was significantly lower than that in sham operation group, while the expression of TGF-β1 in astragalus polysaccharide group was significantly higher than that in sham operation group P <0.01); Western blot method also got the same result. Conclusion Astragalus polysaccharide can play a role in spinal cord injury by up-regulating the expression of TGF-β1.
