目的:优化黄芩黄酮组分的提取工艺,为制备益气活血解毒胶囊奠定基础。方法:以芦丁为对照品,建立黄芩总黄酮含量的测定方法;采用正交试验,以总黄酮得率为指标,考察溶剂量、提取次数、提取时间等对黄芩中总黄酮含量的影响,优选最佳提取工艺。结果:通过对正交试验结果的极差和方差分析,优选黄芩总黄酮提取的最佳工艺为:料液比为1∶10,提取3次,每次2 h。结论:试验结果表明提取次数对黄芩总黄酮的得率有显著影响,其次为提取时间、溶剂量。该工艺简单、方便、合理、可行,适合工业化生产。
Objective: To optimize the extraction process of skullcap flavonoids and lay the foundation for the preparation of capsule for supplementing qi and activating blood circulation and detoxification. Methods: Rutin was used as reference substance to establish the method for determination of total flavonoids in Scutellaria baicalensis Georgi. The effects of solvent amount, extraction times and extraction time on total flavonoids in Scutellaria baicalensis Georgi were studied by orthogonal test. Optimum extraction process is preferred. Results: The optimal extraction process of total flavonoids from Scutellaria baicalensis according to the range and variance analysis of orthogonal test was as follows: the ratio of solid to liquid was 1:10 and extracted three times for 2 h each. Conclusion: The results showed that the extraction times had significant effect on the yield of total flavonoids from Scutellaria baicalensis, followed by extraction time and amount of solvent. The process is simple, convenient, reasonable and feasible, suitable for industrial production.
