目的利用损伤性肾匀浆体外模拟急性肾损伤的微环境,探讨骨髓间充质干细胞(BMSC)诱导转分化及其可能机制。方法将体外培养的BMSC用10%大鼠缺血再灌注损伤肾匀浆上清液进行培养。7d后从形态学改变、18型角蛋白(CK-18)、肝细胞生长因子(HGF)、骨形态发生蛋白-7(BMP-7)的相应表达水平。结果损伤肾匀浆诱导7d后,骨髓间充质干细胞形态发生明显变化,并检测到CK-18阳性表达;同时被诱导的HGF和BMP-7表达水平明显升高。结论损伤肾匀浆可诱导BMSC部分向肾小管上皮样细胞转分化,其可能机制是BMSC可能以旁分泌的形式产生了HGF和BMP-7等“肾脏保护因子”。
OBJECTIVE: To study the microenvironment of acute renal injury (MNC) induced by BMSC in vitro and its possible mechanism. Methods BMSCs cultured in vitro were cultured in 10% of rat renal homogenate supernatant after ischemia-reperfusion injury. The morphological changes, the expression levels of CK-18, HGF and BMP-7 were observed after 7 days. Results After 7 days, the morphology of bone marrow mesenchymal stem cells changed significantly and the positive expression of CK-18 was detected. Meanwhile, the expression of HGF and BMP-7 was significantly increased. Conclusion Damaged renal homogenate induces the transdifferentiation of BMSC into tubular epithelial-like cells. The possible mechanism is that BMSC may produce paracrine forms of HGF and BMP-7, such as “renal protective factor”.
