目的评估2种育发剂对中国仓鼠肺细胞(CHL)细胞活力的影响及其诱导染色体畸变的能力。方法在非代谢活化和代谢活化情况下,采用MTT法和体外哺乳动物细胞染色体畸变试验检测2种(编号A和B)育发剂对CHL细胞活力和染色体畸变率的影响。结果 A育发香波在不含S9的系统中,受试物终浓度为0.156μl/ml时细胞存活率为54.9%;在含S9的系统中,受试物终浓度为0.313μl/ml时细胞存活率为52.4%;B育发膏在不含或含S9的系统中,受试物终浓度为5 mg/ml时细胞存活率均超过90%。与对照组比较,A育发香波各剂量组的染色体畸变率差异无统计学意义(不加S9的体系下Pearson Chi-Square值=0.341,P=0.952,在S9体系下的Pearson Chi-Square值=0.687,P=0.876),B育发膏在加S9和不加S9试验体系下高剂量组的染色体畸变率与阴性对照组相比,有显著性差异(不加S9的体系下Pearson Chi-Square值=16.611,P=0.000,在S9体系下的Pearson Chi-Square值=11.971,P=0.01;重复试验不加S9的体系下Pearson Chi-Square值=15.457,P=0.000,在S9体系下的Pearson Chi-Square值=15.341,P=0.000),低、中剂量组的染色体畸变率无显著性差异(不加S9的体系下Pearson Chi-Square值=0.000,P=1.000,在S9体系下的Pearson Chi-Square值=0.687,P=0.709)。结论在本实验条件下,A育发香波无致突变性,B育发膏有致突变性。
Objective To evaluate the effects of two hair growth promoters on the viability of Chinese hamster lung cells (CHL) cells and their ability to induce chromosome aberrations. Methods MTT assay and in vitro mammalian cell chromosome aberration assay were used to detect the effects of two kinds of hair growth promoters (A and B) on the viability and chromosome aberration rate of CHL cells under non-metabolic activation and metabolic activation.
