目的研究过氧化物酶体增殖因子活化受体γ(PPARγ)在垂体腺瘤组织中的表达情况,分析PPARγ与垂体腺瘤激素免疫分型之间的关系。方法选取2002年1月 ̄2005年5月我科手术切除的垂体腺瘤38例,对它们的病理切片作免疫组织化学染色后PPARγ进行阳性细胞计数,以阳性细胞所占百分比进行半定量分析。结果所有肿瘤病理切片中均发现有PPARγ的表达。PPARγ免疫染色阳性细胞的表达率从8% ̄65%不等。PPARγ染色阳性表现为细胞核染成均一棕黄色至深棕色,主要定位于细胞核内,在细胞浆内也有弱表达。按照垂体腺瘤激素免疫分型,各组肿瘤表达的PPARγ阳性细胞等级比在统计学上未发现有显著差异。分析8例侵袭性垂体腺瘤和非侵袭性垂体腺瘤病例之间PPARγ的阳性细胞率,发现也无统计学上的显著差异。结论人类垂体腺瘤中有PPARγ的表达。PPARγ的人工合成配体-TGZs类药物对所有激素类型和侵袭性或非侵袭性垂体腺瘤都有潜在的治疗效应。
Objective To study the expression of peroxisome proliferator - activated receptor γ (PPARγ) in pituitary adenoma tissue and to analyze the relationship between PPARγ and the hormone immunophenotype of pituitary. Methods Thirty-eight cases of pituitary adenoma resected in our department from January 2002 to May 2005 were enrolled. The positive cells of PPARγ were counted by immunohistochemical staining for their pathological sections. Semi-quantitative analysis was performed on the percentage of positive cells. Results The expression of PPARγ was found in all tumor pathological sections. The expression of PPARγ immunostaining positive cells varied from 8% to 65%. The positive staining of PPARγ was that the nuclei were stained to a homogeneous brown to dark brown color, mainly located in the nucleus and weakly expressed in the cytoplasm. According to the pituitary adenoma hormone immunophenotyping, the rank of PPARγpositive cells expressed by tumors in each group was not significantly different statistically. Analysis of 8 cases of invasive pituitary adenoma and non-invasive pituitary adenoma cases between PPARγ positive cell rates and found no statistically significant difference. Conclusion There is PPARγ expression in human pituitary adenoma. Synthetic ligands for PPARγ, the TGZs, have potential therapeutic effects on all hormone types and on invasive or non-invasive pituitary adenomas.
