目的分析不同浸润类型的胃癌患者幽门螺旋杆菌(Helicobacter pylori,HP)感染情况、病灶组织中环氧酶-2(cyclo-oxygen-ase-2,COX-2)和T淋巴瘤侵袭转移诱导因子(T-lymphomin vasion and metastasis 1,Tiam 1)表达情况以及分析HP感染情况与病灶组织COX-2和Tiam 1表达的相关性。方法 94例胃癌患者纳入本研究,且以正常患者24例为对照组。苏木精-伊红染色法(hematoxylin-eosin staining,HE)和血清HP抗体检测来评价HP的感染情况,用荧光定量聚合酶链式反应、蛋白质印迹法和免疫组化来分析病灶组织,两个结果最终进行回归分析,得出COX-2和Tiam 1与HP感染的相关性。结果胃癌T1患者平均HP抗体浓度为(10.21±2.11)μg/ml,胃癌T2患者平均HP抗体浓度为(19.09±3.61)μg/ml,胃癌T3患者平均HP抗体浓度为(26.82±2.04)μg/ml,胃癌T4患者平均HP抗体浓度为(34.51±4.21)μg/ml。随着血清HP抗体浓度的增加,患者病灶组织的Tiam 1 mRNA、蛋白表达量和IOD值逐渐上升。COX-2和Tiam 1蛋白表达量与血清HP抗体浓度呈正相关性(酌2=0.7485和酌2=0.8084)。结论 HP抗体浓度与COX-2或Tiam1的蛋白表达量具有正相关性,HP抗体浓度检测、COX-2和Tiam 1表达检测具有潜在的评价胃癌浸润程度的临床价值。
Objective To analyze the infection status of Helicobacter pylori (HP) in gastric cancer patients with different infiltration types, the expression of cyclooxygenase-2 (COX-2) and T lymphoma invasion and metastasis inducing factor T-lymphomin vasion and metastasis 1, Tiam 1) and analyze the correlation between HP infection and the expression of COX-2 and Tiam 1 in focal tissue. Methods 94 cases of gastric cancer patients were included in this study, and 24 cases of normal patients as the control group. HP infection was assessed by hematoxylin-eosin staining (HE) and serum HP antibody detection, and the lesions were analyzed by fluorescence quantitative polymerase chain reaction, Western blotting and immunohistochemistry. Results were finally analyzed by regression analysis, and the correlation between COX-2 and Tiam 1 and HP infection was obtained. Results The average HP antibody concentration in gastric cancer T1 was (10.21 ± 2.11) μg / ml, that in gastric cancer T2 was (19.09 ± 3.61) μg / ml, and that in gastric cancer T3 was (26.82 ± 2.04) μg / ml, the average HP antibody concentration in gastric cancer T4 was (34.51 ± 4.21) μg / ml. With the increase of serum HP antibody concentration, Tiam 1 mRNA, protein expression and IOD value gradually increased in the lesion tissues. The expression levels of COX-2 and Tiam 1 protein were positively correlated with serum HP antibody concentration (P = 0.7485 and P = 2, 0.8084). Conclusion The HP antibody concentration has a positive correlation with the protein expression of COX-2 or Tiam1. The detection of HP antibody concentration and the expression of COX-2 and Tiam 1 have the potential clinical value of evaluating the degree of gastric cancer infiltration.
