为了进一步证实EB病毒与干燥综合征发病的关系,作者设计了EB病毒BamW片段特异的引物和寡核苷酸探针,利用PCR体外基因扩增方法和核酸分子杂交技术检测出干燥综合征患者外周血白细胞及唇腺组织中存在有高频率的EB病毒DNA,而对照组的阳性率相对很低。用原位杂交方法检测干燥综合征患者唇腺组织中EB病毒DNA,共28例中有16例为阳性。结果说明干燥综合征患者EB病毒感染率较正常人明显增高,且EB病毒感染与靶器官的损害有关,提示靶器官控制EB病毒细胞内复制的免疫机制出现异常。
In order to further confirm the relationship between Epstein-Barr virus and Sjogren’s syndrome, we designed the BamW fragment-specific primers and oligonucleotide probes of Epstein-Barr virus (EBV) to detect the periplasm of patients with Sjogren’s syndrome using PCR in vitro gene amplification method and nucleic acid molecular hybridization High frequency of Epstein-Barr virus DNA was present in leukocytes and labial glands, while the positive rate in the control group was relatively low. In situ hybridization method for detecting Epstein-Barr virus DNA in the labial gland tissues of Sjogren’s syndrome was positive in 16 out of 28 cases. The results show that patients with Sjogren’s syndrome EB virus infection rate was significantly higher than normal, and the EB virus infection and target organ damage, suggesting that target organs control EB virus intracellular replication of the immune system abnormalities.
