目的:比较大花红景天(Radix et Rhizoma Rhodiolae crenulatae)水提剂与红景天苷(Salidroside)对低氧条件下ECV304细胞表达HIF-1α和VEGF的不同影响。方法:细胞分4组:正常氧对照组,低氧对照组,红景天水提剂组,红景天苷组。采用TaqMan探针实时定量聚合酶链式反应(PCR)方法检测mRNA表达水平;Western Blot法检测蛋白表达水平。结果:与正常氧对照相比,低氧促进ECV304细胞HIF-1α和VEGF基因和蛋白表达(P<0.05);同在低氧条件下,红景天水提剂显著促进了ECV304细胞HIF-1α和VEGF的基因和蛋白表达(P<0.01),而红景天苷的促进作用较弱,没有提高低氧时HIF-1α和VEGF的蛋白水平。结论:红景天促进HIF-1α、VEGF表达时,红景天苷不是主要有效成分。
OBJECTIVE: To compare the different effects of aqueous extract of Radix et Rhizoma Rhodiolae crenulatae and Salidroside on the expression of HIF-1α and VEGF in ECV304 cells under hypoxic conditions. Methods: The cells were divided into 4 groups: normal oxygen control group, hypoxic control group, Rhodiola extract group, salidroside group. TaqMan probe real-time quantitative polymerase chain reaction (PCR) method to detect mRNA expression levels; Western Blot test protein expression levels. Results: Compared with normal oxygen control, hypoxia increased the expression of HIF-1α and VEGF in ECV304 cells (P <0.05). Under hypoxic conditions, Rhodiola rosea significantly promoted the expression of HIF-1α and VEGF in ECV304 cells VEGF gene and protein expression (P <0.01), while the salidroside promotion effect is weak, did not increase the hypoxia HIF-1α and VEGF protein levels. Conclusion: Salidroside is not the main active ingredient when Rhodiola promoted the expression of HIF-1α and VEGF.
