本研究就转录组数据分析得到一个与镉胁迫相关的基因GhHMP1 (Heavy metal transport/detoxification superfamily protein1),并从陆地棉耐镉品种'邯242'中克隆成功.为研究该基因的功能和特点,对其进行生物信息学分析、转录组分析和实时荧光定量表达分析.结果表明:陆地棉'邯242'基因GhHMP1的CDS全长为402 bp,编码133个氨基酸,分子量约为14.88 kD,等电点为8.20;转录组和实时荧光定量分析表明, GhHMP1基因的表达具有组织特异性,且受镉胁迫诱导,可以作为重要的候选基因来培育棉花耐镉新材料,从而为棉花耐镉育种及修复重金属污染土壤提供重要的理论依据.“,”In this study, a gene named GhHMP1 (Heavy metal transport/detoxification superfamily protein 1) associated with cadmium stress was obtained by transcriptome data, and was successfully cloned from cadmium-tolerance variety 'Han242' (Gossypium hirsutum L.). To study the function and characteristics of this gene, the bioinformatics analysis, transcriptome analysis and qRT-PCR analysis were carried out. The results showed that the gene length of GhHMP1 of 'Han242' was 402 bp, encoding 133 amino acids, with the molecular weight of 14.88 kD and the isoelectric point of 8.20. Transcriptome and real-time fluorescence quantiative PCR analysis indicated that the expression of gene GhHMP1 had tissue specificity, and it was induced by cadmium stress. Therefore, it could be used as an important candidate gene to cultivate new cadmium resistant materials, so as to provide important theoretical basis for the breeding of cadmium resistant cultivar and repair of heavy metal contaminated soil.
