目的:建立反相高效液相色谱法检测血浆中坎地沙坦含量。方法:采用Europspher-100C18柱,以甲醇-0.02mo·lL-1磷酸二氢钾缓冲液(66:34,磷酸调pH至2.4)为流动相,缬沙坦作内标,荧光检测器激发波长为265nm,发射波长为395nm。结果:血浆中坎地沙坦浓度在2.23~557.50ng·ml-1范围内线性关系良好,r=0.9999(n=7),低、中、高3个浓度样品的提取回收率和方法回收率分别为57.84%,55.82%,55.23%和91.94%,93.37%,96.42%。结论:本法操作简便,灵敏度高,重现性好,可用于血浆中坎地沙坦的含量测定。
Objective: To establish a method for the determination of candesartan in plasma by reversed-phase high performance liquid chromatography. Methods: Europspher-100C18 column was used with methanol-0.02 mol · L-1 potassium dihydrogen phosphate buffer (66:34, adjusted to pH 2.4 with phosphoric acid) as the mobile phase. Valsartan was used as the internal standard. The fluorescence excitation wavelength Is 265 nm, the emission wavelength is 395 nm. Results: The linearity of candesartan in plasma ranged from 2.23 to 557.50 ng · ml-1, r = 0.9999 (n = 7). The recoveries and recoveries of low, medium and high concentration samples Respectively 57.84%, 55.82%, 55.23% and 91.94%, 93.37% and 96.42%. Conclusion: This method is simple, sensitive, reproducible, and can be used for the determination of candesartan in plasma.
