Mitsunobu反应由于反应条件温和、立体选择性好,在有机合成及药物化学研究中被广泛应用.同时,由于氧化膦及肼类等副产物的影响,该反应也是有机合成化学中最难纯化的反应之一.简化分离、提高纯化效率是近年来对Mitsunobu反应改进研究的热点方向.综述了近年来各种简化分离的Mitsunobu反应,重点介绍了各种分离策略的原理、优缺点及在Mitsunobu反应中的应用.希望通过对Mitsunobu反应各种分离策略的介绍,加深读者对基于分离策略的有机合成的了解.
Mitsunobu reaction is widely used in organic synthesis and medicinal chemistry due to its mild reaction conditions and good stereoselectivity, and is also the most difficult reaction to purify in organic synthesis chemistry due to the influence of by-products such as phosphine oxide and hydrazine I. Simplification of separation and improvement of purification efficiency are the hot topics in the Mitsunobu reaction improvement in recent years.In this paper, we review the Mitsunobu reactions with various simplifications in recent years, highlighting the principles, advantages and disadvantages of various separation strategies and their applications in the Mitsunobu reaction And hope that readers can deepen their understanding of organic synthesis based on separation strategy by introducing various separation strategies of Mitsunobu reaction.
