对新生乳鼠肺、肝来源的原代混合培养细胞,在每ml培养液中加入0.1μci的~3HTdR进行诱导。受~3HTdR作用组细胞能长期存活,接种于免疫抑制动物形成纤维肉瘤,证明发生了恶性转化。埘恶性转化细胞进行染色体分析,见有长臂染色体、中着丝点染色体等稳定型畸变,以及染色体断片、单体断裂等非稳定型畸变,说明~3HTdR对DNA的损伤,是导致细胞染色体畸变,进而发生恶性转化的重要原因。非稳定性畸变在多次传代的转化细胞中仍然存在,说明微量~3HTdR可能残留于DNA中,持续对细胞造成损伤。
