目的 :研究99m Tc标记的大肠癌单克隆抗体(99mTc ACCmAb)在正常小鼠和荷瘤裸鼠体内的分布及在荷瘤裸鼠体内SPECT成像的研究。方法 :将99m Tc和99mTc ACCmAb分别注射入正常小鼠、荷L0 10 16人结肠癌裸鼠 ,按不同时间点采取小鼠的脏器进行同位素放射性活度的测定 ;并在不同时间测定荷L0 10 16人结肠癌裸鼠注射99mTc ACCmAb的SPECT显像。结果 :注射99mTc后正常小鼠体内 6h 5min血液中的放射性活度与 30min相比下降了 76 .6 % ,是30min的 2 3.4 % ;肝脏中 6h 5min的放射性活度为30min的 2 7.2 % ,下降了 72 .8%。尾静脉注射99mTc ACCmAb的正常小鼠 ,6h 5min在血液中的放射性活度与 30min血液中的放射性活度比较下降了6 8.4 % ,是 30min的 31.6 % ,2 4h为 6h 5min的6 .1% ,下降了 93.9% ;荷瘤裸鼠体内99mTc ACCmAb在 2 4h血液中的放射性活度是 6h 5min的 2 .1% ,下降了 97.9%。SPECT显像结果表明 ,虽然在注射99mTc ACCmAb 2 0h血液与肿瘤的比值为最高 ,但在 2 4h的成像结果仍然好于其他的时间点。结论 :99m Tc标记的大肠癌单克隆抗体可特异性浓聚于荷L0 10 16人结肠癌裸鼠模型中 ,可能成为大肠癌根除手术中使用的手术导向体内诊断用药 ,保证手术切除肿瘤的彻底性
AIM: To investigate the distribution of 99m Tc-labeled monoclonal antibody (99mTc-ACCmAb) in normal mice and tumor-bearing nude mice and SPECT imaging in tumor-bearing nude mice. Methods: 99m Tc and 99m Tc ACCmAb were injected into normal mice and L1016 human colon cancer nude mice, respectively. The radioactivity of the mice was measured at different time points. At different time points, L0 SPECT imaging of 99mTc ACCmAb in 10 16 human colon cancer nude mice. Results: The radioactivity of 6h 5min in normal mice after injection of 99mTc decreased by 76.6% compared with 30min, which was 23.4% after 30min. The radioactivity of 6h 5min in liver was 7.2% at 30min, A decrease of 72.8%. The normal mice with tail vein injection of 99mTc ACCmAb had a 6 8.4% decrease in radioactivity in the blood at 6h 5 min compared with 30 min in the blood, 31.6% at 30 min and 6.1% at 24 h 6 h 5 min , Decreased by 93.9%. The radioactive activity of 99m Tc ACCmAb in 24 h blood in tumor-bearing nude mice was decreased to 97.9% at 2.1% of 6 h 5 min. The results of SPECT imaging showed that although the ratio of blood to tumor was highest at 20 min after injection of 99mTc ACCmAb, the imaging results at 24 h were still better than those at other time points. CONCLUSION: The 99m Tc-labeled monoclonal antibody of colorectal cancer can specifically bind to the L01016 human colon cancer model in nude mice and may be used as a surgery-guided in vivo diagnostic drug for the treatment of colorectal cancer and ensure the complete removal of the tumor Sex
