目的:探讨E2F1在肾透明细胞癌中的表达及意义。方法:通过实时定量PCR和Western blot方法检测E2F1在肾透明细胞癌及对应瘤旁组织中mRNA和蛋白的表达情况,并分析E2F1的mRNA水平与临床病理资料的关系及表达相关性。结果:与对应瘤旁组织相比,E2F1的mRNA水平在肾透明细胞癌手术标本中表达明显升高(P=0.0002),相应的蛋白水平对比与mRNA变化一致;E2F1的mRNA水平在不同年龄、性别组间差异无统计学意义(P>0.05),而在组织学分级、肿瘤直径大小、T分期、临床分期和大血管浸润与否差异有统计学意义(P<0.05)。结论:E2F1表达上调可能在肾透明细胞癌的肿瘤形成中发挥作用,且E2F1表达升高可能促进肾透明细胞癌的恶性进展。
Objective: To investigate the expression of E2F1 in renal clear cell carcinoma and its significance. Methods: The mRNA and protein expression of E2F1 in renal clear cell carcinoma and corresponding paracancerous tissues were detected by real-time PCR and Western blot. The relationship between E2F1 mRNA and clinicopathological data and the expression of E2F1 were analyzed. Results: The mRNA level of E2F1 was significantly higher in the specimens of renal clear cell carcinoma (P = 0.0002) than that in the corresponding paracancerous tissues. The corresponding protein level was consistent with that of mRNA. The mRNA level of E2F1 was significantly different at different ages, There was no significant difference between sex groups (P> 0.05), but there was significant difference in histological grade, tumor diameter, T stage, clinical stage and major vascular invasion (P <0.05). Conclusion: The up-regulation of E2F1 may play a role in the tumorigenesis of renal clear cell carcinoma, and the increased expression of E2F1 may promote the malignant progression of renal clear cell carcinoma.
