目的 :建立人膀胱癌细胞系BC 6 ,为膀胱癌的基础研究提供实验模型 .方法 :1 8例膀胱癌标本采用组织块直接培养法、8例采用细胞悬液培养法行原代培养 ;其中 1 2例同时行裸鼠皮下移植 ,在皮下生长后 ,连续裸鼠体内传代 3次 ,再行体外培养 .结果 :组织块直接培养和细胞悬液培养均未成功 .1 2例裸鼠皮下接种肿瘤有 3例生长 ,l例裸鼠意外死亡 ,l例裸鼠体内传代F2代肿瘤消失 .l例肿瘤裸鼠皮下接种后生长较快 ,裸鼠体内传代 3次后 ,取部分肿瘤组织体外培养得到膀胱癌细胞系BC 6 ,其形态结构 ,分化程度与原发瘤保持一致 ,染色体形态仍为人类核型 ,众数维持在 6 6~ 72之间 ,占82 % ,细胞周期分析G1 期 0 .5 8,G2 期 0 .0 8,S1 期 0 .35 ,细胞群体培增时间为 4 4h .结论 :通过裸鼠体内移植瘤建立的膀胱癌细胞系BC 6 ,与原发癌保持相同的生物学特性 ,体外连续传代1a以上 ,传代 92代 .细胞形态不变 ,生长周期恒定 ,己经成为一个稳定的细胞系
OBJECTIVE: To establish a human bladder cancer cell line BC 6 to provide an experimental model for the basic research of bladder cancer.Methods: Twenty-eight bladder cancer specimens were directly cultured by tissue culture and 8 were cultured by cell suspension culture. The chromosome shape was still human karyotype. The number of BC 6 remained between 66 and 72%, accounting for 82%. Cell cycle analysis G1 0 .5 8, G2 phase 0. 08, S1 phase 0 .35, cell population incubation time 4 4 h.Conclusion: The bladder cancer cell line BC 6 established by transplanted tumor in nude mice remains the same as the primary cancer Biological characteristics, continuous passage in vitro more than 1a, passage 92 generation The same cell shape, constant growth cycle, has become a stable cell line
