目的:探讨p38反义寡核苷酸(p38 anti-ODN)对卵巢癌细胞株(CAOV3)增殖的影响。方法:培养卵巢癌细胞株(CAOV3),以MTT比色法及克隆形成实验测定p38 anti-ODN抑制卵巢癌细胞株CAOV3增殖的作用。免疫印迹法测定p-p38、Cyclin D1及p21waf/cip1表达。免疫沉淀法纯化蛋白并p38试剂盒测定p38活性。结果:MTT实验及克隆分析法均显示p38 anti-ODN明显抑制CAOV3细胞增殖、p-p38表达、下调Cyclin D1表达,同时上调p21waf/cip1表达。结论:p38反义寡核苷酸可以抑制卵巢癌细胞株(CAOV3)增殖,其机制与影响p38通路及下游CyclinD1、p21waf/cip1表达有关;干预p38通路是基因治疗卵巢癌的重要途径之一。
Objective: To investigate the effect of p38 antisense oligonucleotide (ODN) on the proliferation of ovarian cancer cell line (CAOV3). Methods: The ovarian cancer cell line (CAOV3) was cultured and the effect of p38 anti-ODN on the proliferation of ovarian cancer cell line CAOV3 was determined by MTT assay and colony formation assay. The expressions of p-p38, Cyclin D1 and p21waf / cip1 were detected by Western blot. The protein was purified by immunoprecipitation and p38 activity was determined by the p38 kit. Results: MTT assay and clone analysis showed that p38 anti-ODN significantly inhibited the proliferation of CAOV3 cells and the expression of p-p38, down-regulated the expression of Cyclin D1 and up-regulated the expression of p21waf / cip1. Conclusion: p38 antisense oligonucleotide can inhibit the proliferation of ovarian cancer cell line (CAOV3), and its mechanism is related to the influence of p38 pathway and the expression of CyclinD1 and p21waf / cip1. Intervention of p38 pathway is one of the important ways of gene therapy for ovarian cancer.
