目的:建立丹红注射液中迷迭香酸在大鼠血浆中浓度的测定方法。方法:雄性SD大鼠6只,静脉注射丹红注射液(10 mL.kg-1)后,分别于不同时间取血,以肉桂酸为内标,液液萃取处理后,HPLC测定浓度。Diamonsil(钻石)C18色谱柱(4.6 mm×250 mm,5μm),乙腈-0.4%的磷酸水为流动相,梯度洗脱,流速1.0 min.mL-1,检测波长286 nm。结果:迷迭香酸在4.0~100.0 mg.L-1呈良好线性关系(R2=0.996 5);回收率在85.0%~101.4%,方法的日内、日间精密度(RSD)分别为4.00%~6.76%,4.50%~7.81%。不同时间点血浆中迷迭香酸的质量浓度分别为85.44,75.09,61.13,54.73,43.02,20.92,10.12,7.18,4.28 mg.L-1。结论:建立的方法灵敏度好、准确率高,可用于丹红注射液中迷迭香酸的药代动力学研究。
Objective: To establish a method for the determination of the concentration of rosmarinic acid in rat plasma by Danhong injection. Methods: Six male Sprague Dawley rats were injected intravenously with Danhong injection (10 mL · kg-1). Blood was drawn at different times. Cinnamic acid was used as internal standard. After the liquid-liquid extraction, the concentration was determined by HPLC. Diamonsil (C18) column (4.6 mm × 250 mm, 5 μm), acetonitrile-0.4% phosphoric acid water was used as the mobile phase and gradient elution was performed at a flow rate of 1.0 min.mL-1 with a detection wavelength of 286 nm. Results: Rosmarinic acid showed a good linearity (R2 = 0.996 5) in the range of 4.0-100.0 mg · L-1 and the recoveries ranged from 85.0% -101.4%. The intra-day and inter-day precision of the method was 4.00% ~ 6.76%, 4.50% ~ 7.81%. The concentration of rosmarinic acid in plasma at different time points were 85.44, 75.09, 61.13, 54.73, 43.02, 20.92, 10.12, 7.18 and 4.28 mg.L-1, respectively. Conclusion: The established method has good sensitivity and high accuracy and can be used to study the pharmacokinetics of rosmarinic acid in Danhong injection.
