目的:测定不同产地冷水花中总黄酮的含量并评价其抗氧化活性。方法:在单因素试验基础上,以总黄酮提取量为指标,选择料液比、提取温度、提取时间为考察因素,采用响应面法优化冷水花总黄酮的提取工艺,测定不同产地冷水花样品中总黄酮的含量并考察其抗氧化活性。结果:最佳提取工艺为料液比1∶19,提取温度71℃,提取时间126 min。冷水花总黄酮提取量7.08 mg·g-1;不同产地样品中总黄酮质量分数4.35~7.10 mg·g-1,具有明显的地域差异性。冷水花总黄酮对1,1-二苯基苦基苯肼和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基的清除能力与维生素C相当。结论:不同产地冷水花中总黄酮具有较好的抗氧化能力,且该能力与总黄酮含量具有明显量效关系。
OBJECTIVE: To determine the content of total flavonoids in Chrysanthemum from different areas and evaluate its antioxidant activity. Methods: On the basis of single factor experiment, taking total flavonoid extraction as index, selecting the ratio of solid to liquid, extraction temperature and extraction time as the investigation factors, the extraction process of total flavonoids in Chrysanthemum morifolium was optimized by response surface methodology, In the total flavonoids content and investigate its antioxidant activity. Results: The optimum extraction process was solid-liquid ratio 1:19, extraction temperature 71 ℃ and extraction time 126 min. The total flavonoids content of Chrysanthemum morifolium was 7.08 mg · g-1, and the mass fraction of total flavonoids from different areas was 4.35-7.10 mg · g-1, which showed obvious regional differences. Scavenging capacity of total flavonoids from Chrysanthemum morifolium on the free radicals of 1,1-diphenyl picrotylhydrazine and diammonium 2,2-azide-bis (3-ethyl-benzothiazole-6-sulfonate) C quite. CONCLUSION: The total flavonoids from Chrysanthemum indicum L. from different areas have good antioxidation ability, and the capacity has obvious dose-effect relationship with the total flavonoids content.
