目的研究甲基强的松龙(MP)上调热休克蛋白(Hsp27)蛋白对受损中枢神经元自我保护及修复机制的激发和增强作用,为进一步促进受损中枢神经元存活和纤维再生创造有利的条件。方法建立中枢神经损伤模型(视神经眼眶内切断术),经或未经MP治疗的术后动物分别存活4、7 14、21、28 d,取视网膜,切片,Nissl染色观察视网膜节细胞(RGCs)形态和存活数量的变化,用免疫组织化学染色的方法观察节细胞层中Hsp27的表达(检测A值)。用SAS软件对数据进行统计学处理。结果MP治疗组可见受损RGCs在71、4 d的存活数量增多,有统计学意义(P<0.01),在受损RGCs表达增多的Hsp27表达更加强烈(检测A值)。RGCs存活率与A值在各个时间点的变化趋势相似。结论MP可使具有保护作用的Hsp27表达上调,从形态学角度提示了MP可能激发并增强受损中枢神经的自我保护及修复机制。
Objective To investigate the effects of MP on the self-protection and repair mechanism of damaged central neurons, and to promote the survival and regeneration of damaged CNS neurons conditions of. Methods The central nervous system injury model (optic orbital septal surgery) was established. The animals were treated with or without MP for 4, 7, 14, 21, and 28 days respectively. Retinal ganglion cells (RGCs) Morphology and survival changes were observed by immunohistochemical staining Hsp27 expression in the cell layer (detection of A value). Data was statistically processed using SAS software. Results The number of surviving RGCs in injured MP group was significantly increased at 71 and 4 days after MP treatment (P <0.01). The expression of Hsp27 increased in injured RGCs (A value). RGCs survival rate and A value at all time points similar to the trend. Conclusion MP can up-regulate the expression of Hsp27 with protective effect, suggesting that MP may stimulate and enhance the self-protection and repair mechanism of damaged central nervous system from the morphological point of view.
