目的:利用噬菌体表面随机肽展示技术淘选获得牙釉质结合肽。方法:以经过预处理的牙釉质片为模板,通过四轮淘选获得牙釉质结合肽的富集。从第3轮和第4轮产物中分离出总计20条单克隆进行测序及亲和力分析。结果:产物中噬菌体滴度在第2轮产物中得到最大富集之后逐渐下降。所测序的20个单克隆中没有发现共有序列。其中有4个单克隆具有明显的强于WM13噬菌体的牙釉质结合力。结论:四个序列被认为具有较强的牙釉质结合能力。所测序列中未发现明显的共有序列。
OBJECTIVE: To obtain enamel binding peptides by panning using phage display technique. Methods: Enriched enamel binding peptides were obtained by four rounds of panning using pretreated enamel pieces as a template. A total of 20 single clones were isolated from rounds 3 and 4 for sequencing and affinity analysis. Results: The phage titer in the product decreased gradually after the maximum enrichment in the second round of product. No consensus was found in the 20 monoclonal sequences sequenced. Four of them had significantly stronger enamel binding than WM13 phage. Conclusion: Four sequences are considered to have strong enamel binding ability. No significant consensus was found in the sequences tested.
