本研究旨在建立一种快速高效的异丙草胺对映体色谱拆分方法.首次筛选比较了9种涂覆型或键合型纤维素类和直链淀粉类不同手性固定相对异丙草胺对映体的拆分性能,系统考察了醇改性剂种类、比例、流动相流速、柱温等因素对拆分效果的影响,确立在Chiralpak AD-H色谱柱上,以正己烷/异丙醇(V/V,99/1)为流动相,柱温35℃,流速1.0mL/min,异丙草胺对映体可得到最优拆分,分离度为2.41.对异丙草胺对映体在手性固定相上保留和分离的热力学机理进行了初步探讨,发现其手性拆分过程受焓控制.
The aim of this study was to establish a rapid and efficient resolution method for enantiomer chromatographic separation of isopropylallyl.The first screening and comparison of nine kinds of coating or bonding cellulose and amylose with different chiral stationary phase The separation performance of the enantiomers was systematically investigated. The effects of alcohol modifier type, ratio, mobile phase flow rate and column temperature on the resolution were systematically investigated. Chiralpak AD- Isopropylalcohol (V / V, 99/1) was used as the mobile phase at the column temperature of 35 ℃ and the flow rate of 1.0 mL / min. The resolution of propisochlor enantiomers was achieved with a resolution of 2.41. The thermodynamic mechanism of amine enantiomers retention and separation on chiral stationary phases was explored and found that their chiral resolution was controlled by enthalpy.
