目的:运用红外光谱三级鉴别技术和高效液相色谱法(HPLC)对芫花条饮片及其水提物的成分进行研究。方法:采用三级红外技术,对各批饮片及水提物进行分析研究;同时采用HPLC法测定主要药效成分西瑞香素含量。分析整理数据,确定饮片及水提物各自的特征图谱,探讨二者成分的异同。结果:一维红外可以看出,芫花条饮片和提取物分别在1 055,1 073 cm~(-1)(C-O)出现了最大吸收;在二阶导数图谱中,水提物中1 707,1 650,1 624 cm~(-1)(C=O),与西瑞香素(1 709,1 651,1 625 cm~(-1))基本一致;在二维相关红外光谱中看出,芫花条饮片和水提物各自具有3个明显的自动峰。液相含量测定结果显示,提取物中西瑞香素的含量明显高于饮片中含量。与饮片相比,芫花条水提物中香豆素类成分含量升高,表明经过水提取后该类成分达到有效富集。结论:红外三级光谱技术为芫花条饮片及其水提物提供了整体的成分信息,二维红外特征光谱可用于二者的鉴别;液相色谱测定主要活性成分的含量,进一步说明成分的变化。
OBJECTIVE: To study the constituents of Daphne genkwa polio strips and their water extracts by three-stage infrared spectroscopy and HPLC. Methods: The three-stage infrared technique was used to analyze and analyze the slices and water extracts of each batch. The content of cedilanin, the main active ingredient, was also determined by HPLC. Analyze the data to determine the respective characteristics of Pieces and water extracts map to explore the similarities and differences between the two components. Results: The one-dimensional infrared showed that the maximum absorption of 1 055 and 1 073 cm -1 (CO) occurred in the pieces and extracts of Daphne genkwa, respectively. In the second derivative spectra, 1 707 , 1 650,1 624 cm -1 (C = O), which is basically the same as that of sirebuterol (1 709, 1 651, 1 625 cm -1). It can be seen from the two-dimensional correlation infrared spectra , Daphne genkwa pieces and water extracts each have 3 distinct auto peaks. Determination of liquid content showed that the content of cedilamine in the extract was significantly higher than that in the slice. Compared with the Pieces, the content of coumarins in the water extract of Daphne genkwa increased, indicating that after water extraction to achieve effective enrichment of such components. CONCLUSION: The infrared three-stage spectroscopy provides the integral component information for the pieces of Daphne genkwa and its aqueous extracts. The two-dimensional infrared spectrum can be used for the identification of the two components. The content of the main active components is determined by liquid chromatography, Variety.
