目的 :观察柯萨奇 腺病毒受体 (CAR)在不同年龄、性别小鼠心肌组织中的表达情况。方法 :采用免疫组化和Western Blotting技术 ,半定量分析胚胎期 18d ,生后 1d ,4、8、12周雄性小鼠及 4周雌性小鼠心肌组织中的CAR含量。结果 :免疫组化半定量分析示胚胎期 18dCAR表达最高 ,生后 1d(新生鼠 )及 4周龄 (幼年鼠 )表达量依次降低 ,8周龄 (成年鼠 )基本不表达 (P <0 .0 5 ) ,12周龄 (老年鼠 )低表达 ,与 8周龄比较 ,差异有统计学意义 (P <0 .0 1) ,并且 ,4周龄雄性小鼠和雌性小鼠相比 ,CAR表达量雄性高于雌性 (P <0 .0 1) ,Western Blot ting结果示CAR表达变化趋势与免疫组化结果相一致。 结论 :小鼠心肌组织中CAR的表达具有明显的时序性 ,而且性别对CAR的表达有影响。
Objective: To observe the expression of coxsackie adenovirus receptor (CAR) in myocardium of different age and sex. Methods: Immunohistochemistry and Western Blotting were used to semi-quantitatively analyze the content of CAR in cardiac muscle of male and 4-week-old female embryos at embryonic day 18, postnatal day 1, 4, 8 and 12 weeks. Results: Semiquantitative immunohistochemical analysis showed that embryonic 18dCAR expression was the highest, the expression of 1D (newborn rat) and 4-week old (young mouse) decreased gradually after birth, but was not found in 8-week old rats (P <0. 0 5), 12 weeks (old mice), compared with 8 weeks of age, the difference was statistically significant (P <0.01), and 4 weeks old male mice compared with female mice, CAR The expression level of males was higher than that of females (P <0.01). Western Blotting showed that the expression of CAR was consistent with the immunohistochemical results. Conclusion: The expression of CAR in mouse myocardium has obvious sequence, and sex has an impact on the expression of CAR.
