目的探讨人雌激素硫酸基转移酶(hEST)基因与雌激素依赖型子宫内膜腺癌发生、发展关系,为其病因学研究提供有意义的帮助。方法采用Western-Blot及RT-PCR方法分别检测中国医科大学附属盛京医院妇科2005年7月至2007年10月40例子宫内膜腺癌组织、20例正常子宫内膜组织、12例不典型增生子宫内膜组织中hEST蛋白及mRNA表达的相对含量。结果 hEST蛋白及mRNA在正常子宫内膜增殖期、分泌期和萎缩内膜组织中的表达差异显著(P<0.01)。hEST蛋白及mRNA在正常子宫内膜组织、不典型增生子宫内膜组织及子宫内膜腺癌组织中的表达含量逐渐降低,三者比较有差异(P<0.01)。hESTmRNA和蛋白含量与子宫内膜腺癌组织学分级、肌层浸润深度、手术病理分期及淋巴结转移等无关(P>0.05)。子宫内膜腺癌组织中,hESTmRNA和蛋白在ER阳性组织中的含量明显高于ER阴性组织,差异显著(P<0.01)。结论 hEST表达降低或缺失可能与雌激素依赖型子宫内膜腺癌的早期形成有一定的关系,但hEST不能作为子宫内膜腺癌病情程度和预后评价的指标。
Objective To explore the relationship between the human estrogen-sulfatase transferase (hEST) gene and the development and progression of estrogen-dependent endometrial adenocarcinoma, and to provide meaningful help for the study of its etiology. Methods Forty cases of endometrial adenocarcinoma, 20 cases of normal endometrium and 12 cases of atypical endometrial carcinoma from Shengjing Hospital Affiliated to China Medical University from July 2005 to October 2007 were detected by Western-Blot and RT-PCR. The relative content of hEST protein and mRNA expression in endometrial hyperplasia. Results The expression of hEST protein and mRNA in endometrial proliferative phase, secretory phase and atrophic endometrium were significantly different (P <0.01). The expression of hEST protein and mRNA in normal endometrial tissue, atypical hyperplasia endometrial tissue and endometrial adenocarcinoma gradually decreased, with significant difference (P <0.01). There was no correlation between hEST mRNA and protein levels and histological grade, depth of myometrial invasion, pathological stage and lymph node metastasis of endometrial adenocarcinoma (P> 0.05). In endometrial adenocarcinoma, the content of hEST mRNA and protein in ER-positive tissues was significantly higher than that in ER-negative tissues (P <0.01). Conclusion The decrease or deletion of hEST expression may be related to the early formation of estrogen-dependent endometrial adenocarcinoma, but hEST can not be used as an indicator of the severity and prognosis of endometrial adenocarcinoma.
