目的分析CDPA液保存急性白血病患者骨髓对RT-PCR方法检测MUC1基因的影响。方法31例初治急性白血病患者各同时留两份骨髓标本,一份立即分离单个核细胞提取总RNA,一份注入CDPA液血袋4℃保存48h后再分离单个核细胞提取总RNA,分别用RT-PCR方法检测MUC1基因的表达并对结果进行比较。结果两组标本提取的总RNAOD260/280都大于1.8;两组标本的MUC1基因检测结果完全一致。结论4℃下CDPA液保存骨髓48h对采用RT-PCR方法检测MUC1基因的结果无影响,基层医院可用此法保存骨髓送上级医院行基因检测。
OBJECTIVE: To analyze the effect of CDBA on the expression of MUC1 gene in bone marrow of patients with acute leukemia by RT-PCR. Methods Twenty-one patients with newly diagnosed acute leukemia were divided into two groups. One sample was immediately isolated from mononuclear cells and the total RNA was extracted. One sample was injected into CDPA blood bag and stored at 4 ℃ for 48h. Total RNA was extracted from mononuclear cells The expression of MUC1 gene was detected by RT-PCR and the results were compared. Results The total RNAOD260 / 280 extracted from two groups of specimens was more than 1.8. The results of MUC1 gene in two groups of specimens were completely consistent. CONCLUSION: The 48-h bone marrow preservation with CDPA liquid at 4 ℃ has no effect on the detection of MUC1 gene by RT-PCR. The primary hospital can use this method to preserve the bone marrow and send it to the superior hospital for genetic testing.
