目的:采用高效液相色谱与电喷雾四极杆飞行时间质谱仪串联技术(HPLC-ESI-Q-TOF-MS)对天葵子95%乙醇提取部位中的化学成分进行定性分析。方法:采用液质联用色谱方法(LC-MS-MS),C18色谱柱(4.6 mm×150 mm,5μm),以乙腈-0.1%甲酸溶液为流动相梯度洗脱,质谱使用ESI离子源,正离子模式采集数据。质量扫描范围为m/z 50~1 500。结果:通过质谱数据分析,并与相关文献对照,从天葵子95%乙醇提取部位共鉴定出11种化合物,分别为4-(甲氧羰基)-2-氨基丁酸、紫草氰苷、耧斗菜内酯、蝙蝠葛内酯、蝙蝠葛氰苷、菲律宾厚壳树苷、天葵氰苷、橙黄胡椒酰胺、对苯二甲酸二丁酯、胡萝卜苷及邻苯二甲酸-二-2-乙基-己酯。结论:基于HPLC-ESI-Q-TOF技术,通过高效液相色谱分离,质谱测定相对分子质量、裂解数据,文献分析检索,能够快速准确地对天葵子95%乙醇提取部位的化学成分进行分离鉴定,为进一步研究天葵子的药效物质基础提供有力证据。
OBJECTIVE: To qualitatively analyze the chemical constituents in 95% ethanol extract of Nitraria lobata by HPLC-ESI-Q-TOF-MS with high performance liquid chromatography and electrospray ionization time-of-flight mass spectrometry. Methods: LC-MS-MS, C18 column (4.6 mm × 150 mm, 5 μm) was used to elute acetonitrile-0.1% formic acid as mobile phase. ESI ion source was used for mass spectrometry. Positive ion mode data acquisition. Quality scan range of m / z 50 ~ 1 500. Results: A total of 11 compounds were identified from 95% ethanol extracts of Amakusa purpurea by mass spectrometry data analysis and related literature, which were 4- (methoxycarbonyl) -2-aminobutyric acid, Glargine, batogranolide, batarcosin, philippine chentoside, sunflower cyanide, orange yellow pepper amide, dibutyl terephthalate, daucosterol and phthalic acid-di-2 Ethyl-hexyl ester. Conclusion: Based on HPLC-ESI-Q-TOF method, the chemical constituents of 95% ethanol extract of Angelica sinensis can be rapidly and accurately separated by high performance liquid chromatography, mass spectrometry, relative molecular mass, lysis data and literature search and retrieval Identification, in order to further study the sunflower seed material basis to provide strong evidence.
