采用激光镊子拉曼光谱(LTRS)技术对圆红冬孢酵母合成油脂和类胡萝卜素进行定量分析,考察不同碳氮比、碳磷比以及碳硫比对圆红冬孢酵母在细胞水平油脂和类胡萝卜合成能力的影响。测定不同浓度植物油的拉曼光谱,其1751cm-1峰峰高与油脂浓度线性关系良好,相关系数R2可达0.97349,说明激光镊子拉曼光谱法是定量分析油脂的有效手段。以1751cm-1峰和1520cm-1峰峰高定量细胞内油脂和胡萝卜素含量,分析不同样品圆红冬孢酵母细胞的拉曼光谱,结果表明,随着碳氮比、碳磷比和碳硫比的升高,圆红冬孢酵母细胞内油脂含量逐步增加,而类胡萝卜素的含量则显著降低。以上结果说明,激光镊子拉曼光谱技术是实时检测圆红冬孢酵母细胞油脂及类胡萝卜素合成和优化发酵培养基的有效工具。
