目的 初步探讨青春期少女Lepdn受体基因Gln223Arg多态性与体成分、骨密度的关系.方法 从北京地区随机抽取舞蹈专业15~17岁女生60名,以同年龄77名普通中学女生为对照.所有调查对象每天的膳食能量和营养素摄入情况用7天24小时膳食调查表收集.用“一年回顾身体活动调查表”了解其体力活动情况,此外测定所有调查对象的血清lepdn、雌激素水平、骨矿含量、骨密度和体成分.用PCR-RFLP法分析其Leptin受体基因Gln223Arg的多态性.结果 未见这些女生的GIn223Arg基因多态性与体成分间存在相关关系.携带GIn223等位基因的舞蹈专业女生的前臂骨密度明显低于不含该等位基因的个体(P<0.05);且携该基因型别的两组女生在所有检测部位的骨密度也均低于Arg223纯合子个体(P>0.05).结论 Leptin受体基因GIn223Arg多态性可能是影响青春期少女骨密度及峰值骨量获得的一个遗传因素,但这种影响与BMI或体成分无关.此结果还有待进一步验证并揭示其内在机制.“,”Objective To investigate the relationship between leptin receptor(LEPR)gene Gln223Arg polymorphism,body composition,and bone mineral density(BMD)in adolescent girls in Beijing.Methods Sixty girls from a dancing school and 77 girls from a vocational high school,at the age of 15-17 years,were randomly selected for this study.Information on daily energy and nutrient intakes was collected using 7-day 24-hour food records.Physical activity level was assessed with“one-year physical activity questionnaires”.Serum leptin and estrogen levels,bone mass,body mineral density (BMD),and body composition were all measured.The LEPR gene Gln223Arg polymorphism was analyzed by PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism).Results No significant relationship was found between the Gin223Arg polymorphism and body composition.After adjusting for age,BMI,physical activity level,leptin and estradiol level,dancers carrying the Gin223 allele had a significantly lower forearm BMD(P<0.05),and all the Gin carrier showed non-significant trend toward lower BMD at all measured sites than those carrying the Arg223 homozygote.Conclusion Leptin receptor gene Gln223Arg polymorphism might be one of the genetic determinants of BMD and the gain of peak bone mass in adolescent girls,yet this effect might not attributed to BMI or body composition.Further study in more students is needed to replicate these data and to clarify the mechanisms involved.
