目的探讨125I粒子组织间植入治疗小鼠移植性肝癌的疗效及其作用机制。方法建立小鼠皮下移植性H22肝癌模型后行125I粒子组织间植入治疗9~18 d,以10%水合氯醛(0.4 g/kg)腹腔注射麻醉小鼠,测肿瘤大小、血常规,取肿瘤组织做病理切片,bax、bcl-2免疫组化,流式细胞仪分析细胞凋亡率及细胞周期,并与对照组比较。结果125I治疗组所有小鼠均存活,肿瘤缩小至平均直径0.44 cm(与对照组比较,P<0.05);血常规中RBC、plt变化不明显(与正常组比较,P>0.05),HGB、WBC下降明显(与正常组比较,P<0.05)。病理检查治疗组残存肿瘤细胞呈岛状分布,可见大量凋亡细胞,肿瘤中央呈大片凝固性坏死,周围广泛纤维化,有少量淋巴细胞浸润;对照组肿瘤细胞丰富,生长旺盛,间质中有大量淋巴细胞浸润,小血管内可见脱落转移的肿瘤细胞。免疫组化治疗组bax表达增加平均为18.51%(与对照组比较,P<0.05),bcl-2表达减少平均为4.94%(与对照组比较,P<0.05);FCM示治疗12、18 d的细胞凋亡率分别为18.13%、20.92%(与对照组比较,P<0.05),G0/G1期比例分别为37.41%、51.01%(与对照组比较,P<0.05),出现G1期阻滞。结论125I粒子组织间植入治疗可诱导H22肿瘤细胞凋亡,出现G1期阻滞,抑制肿瘤增殖,疗效确切。
Objective To investigate the therapeutic effect and mechanism of 125I seed implantation in mice with transplanted liver cancer. Methods The subcutaneously transplanted H22 hepatocarcinoma model was established and then implanted with 125I seeds for 9-18 days. The mice were anesthetized by intraperitoneal injection of 10% chloral hydrate (0.4 g / kg), and the tumor size and blood routine were measured. The tumor tissue was made into biopsy, bax, bcl-2 immunohistochemistry, apoptosis rate and cell cycle were analyzed by flow cytometry, and compared with the control group. Results All the mice in the 125I treatment group survived, with the tumor shrinking to an average diameter of 0.44 cm (P <0.05 compared with the control group); RBC and plt in the blood routine examination did not change significantly (P> 0.05 compared with the normal group) WBC decreased significantly (compared with the normal group, P <0.05). In the pathological examination group, the residual tumor cells were distributed in the shape of island. A large number of apoptotic cells were observed. Large tumor necrosis was found in the center of the tumor, with extensive fibrosis around and a small amount of lymphocyte infiltration. The tumor cells in the control group were abundant and well- A large number of lymphocytic infiltration, shedding of small blood vessels visible tumor cells. The average increase of bax expression was 18.51% (compared with control group, P <0.05), and the average bcl-2 expression decreased by 4.94% (P <0.05 compared with control group) (P <0.05).
