目的:建立HPLC-ELSD法测定黑乳海参皂苷甲nobiliside A冻干脂质体药物含量以及包封率。方法:采用SB-C18柱;流动相为甲醇-0.5%醋酸二元梯度洗脱;检测器为蒸发光散射检测器;采用微柱离心法分离重建后的nobilisideA脂质体和游离药物。结果:nobiliside A在0.120～1.92μg线性关系良好(r=0.9993);方法平均回收率为99.12%(n=9)。结论:本方法简便快速,结果准确可靠,可用于nobiliside A冻干脂质体含量及包封率的测定。 OBJECTIVE: To establish a method for the determination of drug content and entrapment efficiency of nobiliside A freeze-dried liposomes by HPLC-ELSD. Methods: SB-C18 column was used. The mobile phase was methanol-0.5% acetic acid binary gradient elution. The detector was evaporative light scattering detector. The reconstructed nobiliside liposome and free drug were separated by micro-column centrifugation. Results: The linearity of nobiliside A at 0.120-1.92μg was good (r = 0.9993). The average recovery was 99.12% (n = 9). Conclusion: The method is simple and rapid, accurate and reliable, and can be used for the determination of nobiliside A lyophilized liposome content and encapsulation efficiency.
