为研究补阳还五汤抗脑缺血的有效成分部位及其配伍关系,从补阳还五汤中提取了7 类有效成分部位,采用小鼠双侧颈总动脉和迷走神经结扎法造成脑缺血模型,研究各有效成分部位的作用,以正交实验法分析各有效成分部位的交互作用及有效剂量组合。结果:总生物碱、总多糖、总甙、总甙元和挥发油单独应用均可使小鼠耐缺血时间明显延长,而蛋白质和氨基酸单独应用无明显作用;正交实验表明,蛋白质、氨基酸与其它有效部位之间以及两者之间均无交互作用;三水平正交实验表明,在配伍上,各有效成分部位应取的有效剂量组合为:生物碱为生药0.58 g/kg 体重,多糖为生药11.44 g/kg 体重,甙元为生药8.74 g/kg 体重,总甙为生药14.4 g/kg 体重,挥发油为生药2.5 g/kg 体重。结论:在补阳还五汤中,具有抗脑缺血作用的有效成分部位主要为生物碱、多糖、总甙、甙元和挥发油,它们按一定的比例配伍可发挥较好的作用。
In order to study the effect of Buyang Huanwu Decoction on the active components of cerebral ischemia and their compatibility, 7 types of active components were extracted from Buyang Huanwu Decoction. The cerebral ischemic brain was induced by bilateral common carotid arteries and vagus nerve ligation. The blood model was used to study the effect of each active ingredient, and the interactions and effective dose combinations of each active ingredient were analyzed by orthogonal experiments. Results: The total alkaloids, total polysaccharides, total glycosides, total glycosides and volatile oils alone could significantly prolong the time of ischemia tolerance in mice, but protein and amino acid alone had no significant effect. Orthogonal experiments showed that protein and amino acids were There were no interactions between other effective sites and between the two; three levels of orthogonal experiments showed that in the compatibility, the effective dose combination for each active ingredient should be taken as follows: Alkaloids were 0.58 g/kg body weight for crude drugs. The polysaccharides were crude drug 11.44 g/kg body weight, one yuan was crude drug 8.74 g/kg body weight, total wolfberry was raw drug 14.4 g/kg body weight, and volatile oil was crude drug 2.5 g/kg body weight. Conclusion: In Buyang Huanwu Decoction, the active ingredients with anti-cerebral ischemia are mainly alkaloids, polysaccharides, total peony, aglycone and volatile oils, and they can play a good role by combining certain proportions.
