目的研究加味补肝汤对实验性糖尿病大鼠周围神经病变的防治作用。方法以链脲佐菌素(STZ)诱发糖尿病模型。分别于治疗后4周、8周2个时间点处死大鼠,应用化学比色法测定血清中丙二醛(MDA)的含量;用免疫组织化学法和逆转录-聚合酶链反应(RT-PCR)半定量检测坐骨神经中神经生长因子(NGF)蛋白和mRNA的表达。结果①模型组4周、8周大鼠血清中MDA水平明显高于同期正常对照组(P<0.01),随病程的延长模型组大鼠血清中MDA水平逐渐增加,与同期模型组相比,加味补肝汤能降低血清中MDA水平;②免疫组化及RT-PCR检测显示:在4周、8周模型组坐骨神经中NGF的表达较同期正常对照组明显降低(P<0.01),经加味补肝汤和牛磺酸干预后其表达均较同期模型组明显增强(P<0.05),而加味补肝汤优于牛磺酸,在8周时两组比较差异显著(P<0.05)。结论加味补肝汤对实验性糖尿病大鼠周围神经病变具有一定的防治作用。
Objective To study the preventive and therapeutic effects of Jiaweibugan Decoction on peripheral neuropathy in experimental diabetic rats. Methods Streptozotocin (STZ) induced diabetic model. The rats were sacrificed at 4 and 8 weeks after the treatment, respectively. The level of malondialdehyde (MDA) in the serum was determined by chemical colorimetry. The expression of MDA was detected by immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT- PCR) semi-quantitative detection of nerve growth factor (NGF) protein and mRNA expression in the sciatic nerve. Results ① The level of MDA in serum of model group was significantly higher than that of the normal control group at 4 weeks and 8 weeks (P <0.01). With the prolongation of the course of the model, the level of MDA in the serum of the model group increased gradually. Compared with the model group, Compared with the normal control group, the NGF expression in the sciatic nerve in the model group at 4 weeks and 8 weeks was significantly decreased (P <0.01). After the modified Jiawei Bugan decoction could reduce the MDA level in the serum, Bugangan decoction and taurine treatment significantly increased the expression of TGF-β1 compared with the model group (P <0.05), while Jiaweibugan decoction was better than taurine. There was significant difference between the two groups at 8 weeks (P <0.05). Conclusion Jiaweibugan decoction has certain preventive and therapeutic effects on peripheral neuropathy in experimental diabetic rats.
