目的 :观察大鼠心肌细胞内钙波的影象及钙波在心肌细胞内及细胞间的传播。方法 :采用激光共聚焦显微镜及细胞内钙荧光染色的观测方法。结果 :1在激光共聚焦显微镜下观察了由钙荧光探针 Fluo- 3染色的心房肌及心室肌细胞的影象。 2观察了心肌细胞内钙在某一区域瞬时性增高的现象即钙波在心肌细胞内传播的动态影象 ,钙波的发生与细胞外钙浓度大小有关。3观察了钙波在心肌细胞与细胞间连接处的传播动态影象。结论 :心脏在静息时 (尤其高钙负荷时 )出现钙波是钙信号的一种调节机制。钙波具有在细胞内区域性、瞬时性增高 ,并较快在细胞内传播 ,亦可在细胞间传播。
OBJECTIVE: To observe the image of calcium wave in rat cardiomyocytes and the transmission of calcium wave in and between cardiomyocytes. Methods: The laser confocal microscopy and intracellular calcium fluorescence staining methods. Results: 1 The images of atrial fibroblasts and ventricular myocytes stained with calcium fluorescence probe Fluo-3 were observed under confocal laser scanning microscope. 2 observed intracellular calcium transient increase in a region of the phenomenon that calcium wave in the dynamic transmission of myocardial cells, the occurrence of calcium wave and extracellular calcium concentration related to the size. 3 observed the wave of calcium in the myocardial cells and cell connections at the dynamic image transmission. CONCLUSIONS: The presence of calcium in the heart at rest (especially at high calcium loading) is a regulatory mechanism of calcium signaling. Calcium wave in the intracellular region, the transient increase, and spread faster in the cell, can also be transmitted between the cells.
