目的检测用猪Ⅱ型胶原免疫新西兰大白兔的细胞免疫状态。方法用Ⅱ型胶原免疫新西兰大白兔60天,定期抽取血浆检测抗Ⅱ型胶原抗体;第60天取兔的外周血淋巴细胞,取兔脾细胞、淋巴结分离淋巴细胞,进行体外二次Ⅱ型胶原刺激,检测由此引起的反应性的细胞增殖规津。实验分为二组,第一组加入不同浓度PHA作阳性对照,并测定非特异性免疫;第二组加入不同浓度Ⅱ型胶原,检测特异性免疫。结果正常兔的淋巴细胞在PHA剌激下发生增殖,但对Ⅱ型胶原的第一次剌激不发生增殖,而免疫兔对PHA和Ⅱ型胶原的剌激均能发生显著的增殖。结论表明异种Ⅱ型胶原在一定浓度下,可以引起免疫兔的抗Ⅱ型胶原抗体的升高,并可引起兔脾、外周血淋巴细胞增殖,异种Ⅱ型胶原能在体内引起细胞免疫。
Objective To detect the cellular immune status of New Zealand white rabbits immunized with porcine type Ⅱ collagen. Methods New Zealand white rabbits were immunized with type Ⅱ collagen for 60 days. Blood samples were collected for detection of anti - type Ⅱ collagen antibodies. On the 60th day, the peripheral blood lymphocytes of rabbits were harvested, and splenocytes and lymph nodes were isolated from rabbit spleen cells for secondary collagen Ⅱ Stimulation, detection of the resulting reactive cell proliferation regimen. The experiment was divided into two groups. In the first group, different concentrations of PHA were added as positive control and non-specific immunity was measured. In the second group, different concentrations of type II collagen were added to detect specific immunity. Results Normal rabbits’ lymphocytes proliferated under the stimulation of PHA, but the proliferation of type Ⅱ collagen did not occur in the first stimulation, but the rabbits in immunized rabbits could significantly proliferate PHA and type Ⅱ collagen. The results showed that at a certain concentration of xenogeneic collagen type Ⅱ, the anti-type Ⅱ collagen antibody could be raised in rabbits and the proliferation of lymphocytes in the spleen and peripheral blood could be induced. The xenogeneic collagen type Ⅱ could induce cellular immunity in vivo.
