对我国首次分离的辛德毕斯病毒XJ 16 0病毒株全基因组核苷酸序列进行测定 ,阐明该病毒基因组与国外株的差异 ,了解其分子致病机理。测序结果显示 ,XJ 16 0病毒全基因组除 5′末端帽子结构和 3′末端多聚腺苷酸尾外共 116 2 6个核苷酸 ,编码 3731个氨基酸。非结构基因的编码区长 746 4核苷酸 (6 0nt~ 75 2 3nt) ,编码 2 486氨基酸 ,翻译成四种非结构蛋白 (nonstructuralprotein ,nsp1 4)。结构基因长 3738核苷酸 (75 6 8~ 1130 6 ) ,编码 12 45个氨基酸 ,翻译成三种结构蛋白和两个小分子肽 (capsid ,E1- 3、6k)。病毒基因组 5′末端和 3′末端及结构基因和非结构基因之间分别有 5 9个核苷酸、45个核苷酸和 32 3个核苷酸的非编码区。核苷酸序列分析表明 ,XJ 16 0病毒具备典型的甲病毒属辛德毕斯病毒基因组特征 ,与标准辛德毕斯病毒AR339病毒株相比 ,核苷酸序列存在 18%差异 ,氨基酸差异为 8%。这是我国完成的第一株辛德毕斯病毒全基因组序列测定 ,序列已输入国际基因库 (GenBankAF10 372 8)。
The nucleotide sequence of the whole genome of XJ160 strain of Sindbis virus that was isolated in China for the first time was determined. The difference between the genome of the virus and that of foreign strains was clarified, and the molecular pathogenic mechanism was also studied. Sequencing results showed that the genome of XJ160 virus encoded a total of 116,26 nucleotides in addition to the 5’-end cap structure and 3’-terminal polyA tail, encoding 3731 amino acids. The coding region of nonstructural gene is 746 4 nucleotides (60 nt ~ 75 2 3 nt), encoding 2 486 amino acids, translated into four nonstructural protein (nsp1 4). The structural gene is 3738 nucleotides in length (756 to 11306) encoding 1245 amino acids and translated into three structural proteins and two capsid (E1-3,6k). Between the 5 ’end and 3’ end of the viral genome and the non-coding region of 59, 45 and 32 3 nucleotides, respectively, between the structural and nonstructural genes. Nucleotide sequence analysis showed that the XJ160 virus possesses the typical genus of the alphavirus belonging to Sindbis virus. Compared with the standard Sindbis virus strain AR339, the nucleotide sequence has 18% difference and the amino acid difference is 8%. This is the first complete genome sequence determination of Sindbis virus in our country. The sequence has been entered into the International GenBank (GenBank AF10 372 8).
