目的 通过体外实验研究人体甲状腺乳头状癌细胞对牛 -促甲状腺激素 (TSH)、去甲肾上腺素的反应性 ,阐述它们之间的关系 ,观察甲状腺癌细胞生长的影响因素 ,探讨其内分泌治疗的机理。方法 实验标本来自手术病例 ,病理类型为乳头状癌。修理组织、漂洗、剪碎 ,胰酶和胶原酶 型双酶消化 ,96孔板置于 37℃、5 % CO2 、95 %湿度孵箱中培养。用不同浓度的 b TSH和去甲肾上腺素分别刺激 72 h;非放射细胞增殖法测定细胞生长。结果 显示肿瘤细胞对 b TSH无反应 ;而去甲肾上腺素促进癌细胞生长。结论 b TSH不能促进甲状腺乳头状癌细胞生长 ,提示临床上甲状腺癌的 TSH抑制治疗机理有待进一步探讨 ;另外提示甲状腺癌也受到肾上腺素神经内肽的调节。
Objective To study the response of human papillary thyroid cancer cells to bovine-thyroid-stimulating hormone (TSH) and norepinephrine in vitro, to elucidate the relationship between them, to observe the influencing factors of thyroid cancer cell growth, and to explore the endocrine therapy mechanism. Methods The experimental specimens were from surgical cases. The pathological type was papillary carcinoma. Tissues were repaired, rinsed, and cut into pieces, digested with trypsin and collagenase enzymes, and cultured in 96-well plates at 37°C, 5% CO2, and 95% humidity incubator. After stimulation with different concentrations of b TSH and norepinephrine for 72 h, cell growth was measured by non-radioactive cell proliferation assay. The results showed that tumor cells did not respond to b TSH; norepinephrine promoted cancer cell growth. Conclusion b TSH can not promote the growth of thyroid papillary carcinoma cells, suggesting that the therapeutic mechanism of TSH inhibition in clinical thyroid cancer needs to be further explored; furthermore, thyroid cancer is also regulated by adrenalin-derived neuroendrin.
