目的 观察小鼠胸腺树突状细胞 (TDC)的形态。 方法 密度梯度离心结合小鼠 CD5单抗淘洗法(panning) ,分离出小鼠 TDC并行光镜及电镜观察。 结果 台盼蓝排斥试验示 90 %以上细胞存活。光镜下发现初分离的 TDC常和周围的胸腺细胞形成紧密的细胞簇。S- 10 0蛋白染色见 TDC胞体和突起内有棕黄色反应产物。ANAE反应呈阴性或在核附近有数个小颗粒反应。ATP酶反应阴性。电镜见细胞表面有许多树状突起 ,核形不规则 ,胞质电子密度低 ,有丰富的线粒体、核糖体及完整的高尔基复合体。 结论 所分离的 TDC具有典型的树突状细胞的形态学特征
Objective To observe the morphology of mouse thymus dendritic cells (TDC). Methods Density gradient centrifugation combined with mouse CD5 monoclonal antibody panning, TDC parallel light and electron microscopy were isolated. Results Trypan blue exclusion assay showed that more than 90% of the cells survived. Light microscope revealed that initially isolated TDC often and closely surrounding thymocytes formed tight clusters of cells. S-10 0 protein staining TDC cell body and protrusions have brown-yellow reaction products. The ANAE reaction is negative or there are several small particles near the core. ATP enzyme reaction negative. Electron microscopy see the cell surface has many dendrites, irregular nuclear shape, low cytoplasmic electron density, rich in mitochondria, ribosomes and complete Golgi complex. Conclusion The isolated TDC has typical morphological characteristics of dendritic cells
