目的建立一个瘤细胞标本纯度参照体系以降低标本不纯对抑癌基因杂合性缺失(LOH)研究的影响。方法根据代表不同纯度的标本中两个等位基因的配比推算纯合子/杂合子DNA的构成比及掺入量,进而建立瘤细胞模拟纯度梯度体系,以APC基因的LOH分析为例,在优化实验条件基础上构建不同纯度瘤细胞的APC-LOH模型。结果该APC-LOH分析系统至少能分析纯度低至60%的标本,优于其他文献提出的纯度要求(>70%)。结论本文构建的瘤细胞纯度梯度体系便于优化LOH的实验条件,并能确定优化后实验系统的分辨能力。
Objective To establish a reference system for the purity of tumor cell specimens to reduce the influence of impure specimens on the loss of heterozygosity (LOH) in tumor suppressor genes. Methods According to the ratios of two alleles representing different purity samples, the ratio of the homozygous/heterozygous DNA and the amount of incorporation were calculated. Then the purity gradient system of tumor cells was established. The LOH analysis of APC gene was used as an example. The APC-LOH model of different purity tumor cells was constructed based on optimized experimental conditions. Results The APC-LOH analysis system can analyze samples with purity as low as 60% at least, which is superior to the purity requirements (>70%) proposed by other literatures. Conclusion The purity gradient system of the tumor cells constructed in this study is convenient for optimizing the experimental conditions of LOH and determining the resolving power of the optimized experimental system.
