目的探讨中药白芷各来源的亲缘关系,明确中药白芷的原植物问题。方法用随机扩增多态DNA(RAPD)方法对中药白芷的4种商品类型祁白芷、禹白芷、杭白芷、川白芷和兴安白芷[Angelica dahurica(Fisch.ex Hoffm.)Benth.et Hook.f.ex Franch.etSav.]、台湾白芷(A.formosana de Boiss)、雾灵当归(A.porphyrocaulis Nakai et Kitagawa)及黑水当归(A.amurensis Sehiseh)进行分析,在此基础上,对其ITS区作了序列分析。结果RAPD分析共扩出206条带,其中多态性条带188条,占88.3%,通过计算遗传距离得到严格一致树。各样品扩增所得ITS1序列为439 bp,ITS2序列为410 bp,进行排序和同源性比较,4种商品白芷的序列与台湾白芷是完全相同的。对台湾白芷、兴安白芷、雾灵当归和黑水当归的ITS 1,ITS 2与ITS 1+ITS 2分别进行最大简约树和邻接树的计算,6组数据得到2种树型。结论4种商品类型来源于台湾白芷,建议定名为Angelica formosana de Boiss cv.officinarum cv.nov。雾灵当归、兴安白芷和台湾白芷应作为互相独立的种来对待。
Objective To explore the genetic relationship among the various sources of Radix Paeoniae Alba, and to clarify the original plant problems of Radix Paeoniae Alba. METHODS: Random amplified polymorphic DNA (RAPD) method was used to compare the four commercial types of Radix Paeoniae Alba, namely Radix Paeoniae Alba, Radix Paeoniae Alba, Rhizoma Polygoni, Rhizoma Polygoni and Angelica dahurica [Angelica dahurica (Fisch.ex Hoffm.) Benth.et Hook.f. .ex Franch.etSav.], A.formosana de Boiss, A.porphyrocaulis Nakai et Kitagawa and A.amurensis Sehiseh, based on which ITS The district made a sequence analysis. Results A total of 206 bands were amplified by RAPD analysis, of which 188 bands were polymorphic, accounting for 88.3%. A strict consensus tree was obtained by calculating the genetic distance. The ITS1 sequence obtained from the amplification of each sample was 439 bp, and the ITS2 sequence was 410 bp. Sorting and homology comparisons were performed. The sequences of the four commercial white peony roots were identical to those of Taiwan white peony. For the ITS 1, ITS 2 and ITS 1+ ITS 2 of Taiwan White Pelican, Xingan White Pelican, Wuling Angelica and Blackwater Angelica, the maximum parsimony tree and adjoining tree were calculated respectively. The 6 groups of data were obtained as two tree types. Conclusion The four types of commodities are derived from the Taiwan white pheasant and it is suggested that it be named Angelica formosana de Boiss cv.officinarum cv.nov. Wuling Angelica, Xingan Baiji and Taiwan Baiji should be treated as independent species.
